In 1990, she became the first elected female judge of the Westchester County Court and then became the first female district attorney of Westchester County three years later. Simpson murder trial, appearing on Nightline and Larry King Live.

She attempted to secure the Republican nomination for the state to run against Hillary Clinton for a U.S. Senate seat from New York in 2006. Even Trump contributed to her campaign. Ultimately, she withdrew from the race to instead run for the position of New York State Attorney General in the same year. She lost to Democrat Andrew Cuomo.

Jeanine was married to Albert Pirro, a prominent lawyer and businessman in Westchester County, from 1975 until their divorce in 2013. In 2000, Albert was found guilty on 34 counts of tax fraud and conspiracy for using over $ 1 million in corporate funds to cover personal expenses, including luxury cars and a vacation home. He was sentenced to 29 months in federal prison, serving 11 months before being released early. Despite publicly supporting him during the trial, Jeanine later described this time as a difficult period in her life.
